Mimicking tumor microenvironment to model metastatic Colorectal Cancer (CRC ) patient-derived organoids dynamics

Descrizione This study introduces a novel matrix based on gelatin methacrylyol (GelMA) for enhancing 3D models of metastatic colorectal cancer (CRC). Traditional 2D cell cultures inadequately replicate the complex tumor microenvironment, limiting the accuracy of preclinical drug testing and cancer research. The newly developed GelMA-based matrix aims to overcome these limitations by providing a more physiologically relevant scaffold that mimics the extracellular matrix of metastatic CRC. The GelMA matrix was synthesized and optimized for mechanical properties and biocompatibility, supporting realistic cell morphology, proliferation, and invasion behaviors. In vitro assays demonstrated that the GelMA-based matrix outperforms existing 3D culture systems in maintaining essential cellular functions and interactions. This innovation promises to improve the predictive accuracy of drug testing and deepen the understanding of metastatic mechanisms in CRC, paving the way for more effective therapeutic strategies.
